    Effects of calibration uncertainties on the detection and parameter estimation of isotropic gravitational-wave backgrounds

    Gravitational-wave backgrounds are expected to arise from the superposition of gravitational wave signals from a large number of unresolved sources and also from the stochastic processes that occurred in the Early universe. So far, we have not detected any gravitational wave background, but with the improvements in the detectors' sensitivities, such detection is expected in the near future. The detection and inferences we draw from the search for a gravitational-wave background will depend on the source model, the type of search pipeline used, and the data generation in the gravitational-wave detectors. In this work, we focus on the effect of the data generation process, specifically the calibration of the detectors' digital output into strain data used by the search pipelines. Using the calibration model of the current LIGO detectors as an example, we show that for power-law source models and calibration uncertainties 10%\lesssim 10 \%, the detection of isotropic gravitational wave background is not significantly affected. We also show that the source parameter estimation and upper limits calculations get biased. For calibration uncertainties of 5%\lesssim 5 \%, the biases are not significant (2%\lesssim 2 \%), but for larger calibration uncertainties, they might become significant, especially when trying to differentiate between different models of isotropic gravitational-wave backgrounds.Comment: 11 pages, 7 figure

    Interactions of neuropathogenic Escherichia coli K1 (RS218) and its derivatives lacking genomic islands with phagocytic Acanthamoeba castellanii and nonphagocytic brain endothelial cells

    Here we determined the role of various genomic islands in E. coli K1 interactions with phagocytic A. castellanii and nonphagocytic brain microvascular endothelial cells. The findings revealed that the genomic islands deletion mutants of RS218 related to toxins (peptide toxin, alpha -hemolysin), adhesins (P fimbriae, F17-like fimbriae, nonfimbrial adhesins, Hek, and hemagglutinin), protein secretion system (T1SS for hemolysin), invasins (IbeA, CNF1), metabolism (D-serine catabolism, dihydroxyacetone, glycerol, and glyoxylate metabolism) showed reduced interactions with both A. castellanii and brain microvascular endothelial cells. Interestingly, the deletion of RS218-derived genomic island 21 containing adhesins (P fimbriae, F17-like fimbriae, nonfimbrial adhesins, Hek, and hemagglutinin), protein secretion system (T1SS for hemolysin), invasins (CNF1), metabolism (D-serine catabolism) abolished E. coli K1-mediated HBMEC cytotoxicity in a CNF1-independent manner. Therefore, the characterization of these genomic islands should reveal mechanisms of evolutionary gain for E. coli K1 pathogenicity

    Higher DNA methylation of ABO gene promoter is associated with acute myocardial infarction in a hospital-based population in Karachi

    Objective: To find out if there is any relationship of methylation status of ABO gene promoter with the risk of acute myocardial infarction (AMI) in a hospital-based Pakistani population in Karachi, Pakistan.Methods: A case control study comprising of 39 adult AMI patients (both males and females; age range 30-70 years) and 39 normal healthy controls (both males and females and similar age range) nested in a large study (to see the relationship of ABO genotypes with AMI) was designed to investigate the methylation status of ABO gene promoter and its association with AMI. The study was carried out at the Aga Khan University, Karachi during July 2018 to June 2019. DNA isolated from samples of AMI patients and normal healthy controls were converted into bisulphite DNA using a kit method. Methylation specific polymerase chain reaction was carried out to determine the methylation status of ABO gene promoter in both cases and controls. Logistic regression was used to find out any association between increased methylation status of ABO gene promoter and risk of AMI.Results: A significantly higher percentage of DNA methylation of the ABO gene promoter was observed in AMI patients as compared to normal healthy controls (82.1% vs. 35.9%; p value \u3c0.001). This higher methylation status of ABO gene promoter was associated with AMI and the odds of AMI in this population were more than 6-fold in subjects with methylated gene promoter compared to those with unmethylated gene promoter after adjusting with age and waist circumference [AOR (95% CI) = 6.27 (1.76-22.3); p value = 0.005].Conclusion: The ABO gene promoter\u27s hypermethylation appears to be increasing the risk of AMI in a hospital-based Pakistani population in Karachi, Pakistan

    Knowledge and attitudes about tetanus and rabies: a population-based survey from Karachi, Pakistan

    OBJECTIVE: To evaluate public knowledge regarding predisposing factors, fatality and prevention of Tetanus and Rabies and attitudes toward vaccination and post-exposure prophylaxis. METHODS: A population-based, cross-sectional survey was conducted in all the 18 towns of Karachi, the largest metropolitan city of Pakistan, from December 2007 to January 2008. Men and women of more than 18 years of age were included in the study which used a self-reporting questionarre as its tool. RESULTS: There were 1201 people interviewed by the study. The majority of respondents had known or heard about Tetanus (n = 973; 81%) and rabies (n = 699; 58%). There were 29 (2.5%) reported dog bites on the subjects themselves and 218(18%) respondents reported dog bites among their family members during the preceeding one year. Only three (11%) of these dog bite victims received some kind of vaccine or post-exposure prophylaxis. The majority of the participants were not aware of the fatality of these diseases and the importance of vaccination and post-exposure prophylaxis. Of the total respondents, 563 (47%) reported an injury or wound during the preceeding one year. Of them, 426 (76%) received a Tetanus injection. Out of the total study population, 1019 (85%) respondents did not know that Tetanus could be a fatal disease, and 844 (70%) did not know that Tetanus could affect and kill newborns. Literate people and males were more likely to have adequate knowledge on multivariate analysis. CONCLUSION: Minor injuries and dog bites are a common occurrence in Karachi. Only a small proportion of these patients received post-exposure treatment. Most of the participants were not aware of the fatality of these diseases and the importance and affordability of vaccination in case of dog bites and minor trauma

    A Novel Bandwidth Enhancement Technique for Cascode Amplifier

    In this paper, a new technique is presented to increase the bandwidth for a single stage amplifier. Usually, -3 dB bandwidth of single stage amplifier is in few MHz. High output impedance and subsequent capacitive loading decrease the bandwidth of amplifier. The presented technique uses a load which itself acts as bandwidth enhancer. This high speed amplifier is designed on 180 nm CMOS technology, operates at 2.5 V power supply. This amplifier is succeeded by an output buffer to achieve a better linearity, high output swing and required output impedance for matching

    Anti-Acanthamoebic properties of resveratrol and demethoxycurcumin

    Acanthamoeba is an opportunist protist pathogen that is known to infect the cornea to produce eye keratitis and the central nervous system to produce fatal granulomatous encephalitis. Early diagnosis, followed by aggressive treatment using a combination of drugs is a prerequisite in successful treatment but even then, prognosis remains poor due to lack of effective drugs. The overall aim of the present study was to determine the anti-Acanthamoebic potential of natural compounds, resveratrol and curcuminoids. Adhesion and cytotoxicity assays were performed using primary human brain microvascular endothelial cells, which constitute the blood–brain barrier. Pre-exposure of organisms to 100 μg resveratrol and demethoxy curcumin prevented amoeba binding by 57% and 73%, respectively, while cytotoxicity of host cells was inhibited by 86%. In an assay for viability of amoebae in the absence of host cells, resveratrol and de-methoxy curcumin exhibited significant amoebicidal effects (23% and 25%, respectively) at 100 μg concentrations (P \u3c 0.01). Neither resveratrol nor demethoxycurcumin had any effect on the proteolytic activities of Acanthamoeba castellanii. Of both compounds, resveratrol is of most interest for further investigation, because of the selective toxicity of resveratrol on A. castellanii but not the human brain microvascular endothelial cells

    Analysis and Optimization of Axial Flux Permanent Magnet Machine for Cogging Torque Reduction

    In this paper, a hexagonal magnet shape is proposed to have an arc profile capable of reducing torque ripples resulting from cogging torque in a single-sided axial flux permanent magnet (AFPM) machine. The arc-shaped permanent magnet increases the air-gap length effectively and makes the flux of the air-gap more sinusoidal, which decreases air-gap flux density and hence causes a reduction in cogging torque. Cogging torque is the basic source of vibration, along with the noise in PM machines, since it is the main cause of torque ripples. Cogging torque is independent of the load current and is proportional to the air-gap flux and the reluctance variation. Three-dimensional finite element analysis (FEA) is used in the JMAG-Designer to analyze the performance of the conventional and proposed hexagonal-shaped PM AFPM machines. The proposed shape is designed to reduce cogging torque, and the voltage remains the same as compared to the conventional hexagonal-shaped PM machine. Further, optimization is performed by utilizing an asymmetric overhang. Latin hypercube sampling (LHS) is used to create samples, the kriging method is applied to approximate the model, and a genetic algorithm is applied to obtain the optimum parameters of the machine
